چکیده

Purpose: To investigate the role of centrosomal protein55 (CEP55) in melanoma.
Methods: The CEP55 expression human melanoma cells and normal melanocytes were determined by quantitative reverse transcription-polymerase chain reaction (qRT-PCR) western blot. Cell proliferation was evaluated colony formation CCK8 assays, while cell cycle assessed flow cytometry. Transwell assay used to migration invasion. Results: protein higher A-2058 A375 than HEMn-LP cells. Overexpression increased viability number colonies (p < 0.01), silencing reduced proliferation. promoted 0.01) via decreased p21 expression, but CyclinD1 CDK1 expression. Knockdown enhanced induce arrest. suppressed invasion downregulating p-AKT p-PI3K. Conclusion: Centrosomal functions as an oncogene activating PI3K/AKT signaling. Therefore, might be a promising target for diagnosis treatment.

Methylation and Loss of Secreted Frizzled-Related Protein 3 Enhances Melanoma Cell Migration and Invasion

BACKGROUND Wnt signaling is important in development and can also contribute to the initiation and progression of cancer. The Secreted Frizzled Related Proteins (SFRPs) constitute a family of Wnt modulators, crucial for controlling Wnt signaling. Here we investigate the expression and role of SFRP3 in melanoma. METHODOLOGY/PRINCIPAL FINDINGS We show that SFRP3 mRNA is down-regulated in malign...

YAP1 enhances cell proliferation, migration, and invasion of gastric cancer in vitro and in vivo

Yes-associated protein 1 (YAP1) plays an important role in the development of carcinomas such as breast, colorectal, and gastric (GC) cancers, but the role of YAP1 in GC has not been investigated comprehensively. The present study strongly suggests that YAP1 and P62 were significantly up-regulated in GC specimens, compared with normal gastric mucosa. In addition, the YAP1high P62high expression...

MicroRNA‐520c enhances cell proliferation, migration, and invasion by suppressing IRF2 in gastric cancer

Dysregulation of microRNA (miRNA) is actively involved in the development and progression of gastric cancer (GC). MiR-520c was previously found to be overexpressed in GC specimens and cells. However, the clinical significance of miR-520c and its biological function in GC remain largely unknown. Here, we found that miR-520c expression in GC tissues was significantly increased compared to normal ...

Carvacrol Alleviates Prostate Cancer Cell Proliferation, Migration, and Invasion through Regulation of PI3K/Akt and MAPK Signaling Pathways

TRPM7 is a potential therapeutic target for treatment of prostate cancer. In this study, we investigated the effects of nonselective TRPM7 inhibitor carvacrol on cell proliferation, migration, and invasion of prostate cancer PC-3 and DU145 cells. Our results showed that carvacrol blocked TRPM7-like currents in PC-3 and DU145 cells and reduced their proliferation, migration, and invasion. Moreov...

SOX4 promotes melanoma cell migration and invasion though the activation of the NF-κB signaling pathway

SOX4 has been reported to be abnormally expressed in many types of cancer, including melanoma. However, its role in cell proliferation and metastasis remains controversial. In this study, SOX4 was downregulated or overexpressed in A375, A2058 and A875 melanoma cells by siRNA or lentivirus transfection, respectively. Cell metastasis was observed by Transwell assay. In an aim to elucidate the und...
